Sumario: | The research focused on metabolite analysis is an analytical discipline that is currently gaining much relevance thanks to the large amount of biological information from an organism that it reveals. The increasing knowledge of metabolic and lipid profiles has revealed the prominent role played by some biological compounds in health as well as in the development of various diseases and disorders.
In turn, special care must be taken in the pre-analytical stages and the pretreatment of samples that are carried out in clinical laboratories in the order to show how these procedures can condition and alter the metabolites of biological samples. For this reason, all possible parameters or variables involved both in the pre-analytical stages and the pretreatment of the samples involved must be analyzed since they can introduce inaccuracies and distort the results. It has been observed how various stimuli in the sample manipulation prior to the analysis can trigger an undesirable response in the metabolites, inducing a series of transformations such as degradation, which result in the modification of the chemical characteristics of starting samples. The analysis of these altered samples irreversibly compromises the quality of the results that can be obtained from them.
For this reason, one of the main objectives of this doctoral thesis is to provide clinical laboratories with a series of reliable protocols and methodologies for the pre-analytical and sample pretreatment stages of the analysis, thus minimizing the error bias that can be introduced at these stages. These errors translate in inaccurate and incongruent values with the reality of the biological sample and it is irremediably associated with an imprecise diagnosis of the organism under study. Firstly, emphasis will be placed on the processes involved in the sample thawing stages and liquid-liquid extraction pretreatment.
Furthermore, this work also aims to show how the combined use of analytical methods based on “fingerprint” techniques together with chemometric tools achieve the detection and identification of markers of different stages of the same disease in people who have an altered metabolism. In particular, this doctoral thesis has focused on obtaining the metabolic and lipid profiles through the use of infrared spectroscopy and liquid chromatography in order to distinguish and classify patients with different states of viral disease induced by the human immunodeficiency virus. |
